They wonder, "After a certain period, don't the cells just grow? Can't they even die?" Well, here's the truth: cell culture is not something you can take for granted. It requires strict aseptic techniques and attention to detail. Let’s go through the proper aseptic procedures for cell culture:
1. Before starting any experiment, make sure the sterile room and laminar flow hood are sterilized using a UV lamp for 30–60 minutes. Then, wipe the work surface with 70% ethanol and turn on the fan for 10 minutes to ensure clean air circulation. Only one cell line should be handled at a time, even if the media is the same, to prevent cross-contamination. After the experiment, remove all materials from the hood, wipe the surface again with 70% ethanol, and wait at least 10 minutes before working on another cell line.
2. Keep the aseptic work area clean and organized. Only essential items like pipette racks, straws, or pipette boxes should be placed inside. Remove other lab equipment after use to maintain good airflow. Always wipe your test articles with 70% ethanol before bringing them into the hood. Perform all procedures in the central sterile area, not near the edges where contamination risk is higher.
3. Handle all sterile materials carefully to avoid contamination. Never touch the tip of a pipette or the mouth of a container. Avoid working directly over open containers. When opening a bottle, hold the cap with one hand and the body of the bottle with the other, keeping it at a 45-degree angle. Never place the cap on the table with the opening facing up.
4. Lab personnel must wear lab coats and gloves at all times. For cell lines derived from humans or viruses, use a Class II biosafety cabinet. Be cautious of aerosol generation, toxic substances like DMSO or TPA, and avoid injuries from sharp needles.
5. Regular maintenance checks are essential:
- Check the COâ‚‚ cylinder pressure.
- Monitor the COâ‚‚ incubator for temperature, COâ‚‚ concentration, and water level (use sterile water and replace it weekly).
- Ensure proper airflow in the hood, and replace UV lamps and HEPA filters regularly (pre-filter every 300 hours, HEPA filter every 3000 hours).
6. To keep the sink area clean, add a disinfectant like Zephrin (1:750) to the sink and change the water periodically.
Proper aseptic technique isn’t just about following rules—it’s about protecting your cells, your experiments, and your safety. A small mistake can ruin weeks of work. Stay focused, stay clean, and always prioritize sterility.
