One of the most common challenges in SPE is managing flow rate. Factors like vacuum, pressure, and gravity can all influence how quickly the liquid moves through the SPE column. Maintaining a stable flow rate is essential for effective extraction, especially during sample loading and elution steps. If the flow rate is too fast, it may reduce the recovery of the target compound, particularly in ion-exchange based extractions. On the other hand, a slow flow rate can increase analysis time and lower overall efficiency. To address this, the AutoSPE system uses six high-precision ceramic metering pumps combined with positive pressure loading, ensuring a consistent and accurate flow rate according to the specified sample volume. This helps maintain high accuracy and repeatability in analytical results.
Another critical issue is the recovery rate. When the recovery rate is not satisfactory, it's important to systematically investigate the cause and improve the extraction method. A stable and reliable method is crucial, and controlling the flow rate plays a major role in achieving this.
Pollution is another concern in SPE. Contaminants in the column are often related to the concentration of the target compound and the detection method used. Reputable manufacturers usually ensure contamination levels are kept within parts per billion (ppb). However, if contaminants interfere with the detection of the target compound, pre-cleaning the column is recommended.
